LY 344864 is a 5-HT 1F receptor agonist for neurological disease research

**Background**

The serotonin 5-HT 1F receptor is a G protein-coupled receptor primarily expressed in the central nervous system and on the trigeminal nerve terminals. Due to its specific distribution, the 5-HT 1F receptor has become a significant target for treating neurological disorders, particularly migraine and neurodegenerative diseases. In migraine research, the activation of these receptors is thought to inhibit the release of pro-inflammatory neuropeptides, thereby reducing neurogenic inflammation. Furthermore, recent evidence suggests that 5-HT 1F receptor activation may promote mitochondrial biogenesis, offering potential neuroprotective effects in conditions such as Parkinson’s disease. In this context, we will introduce a selective 5-HT 1F receptor agonist – LY 344864.

**Definition**

LY 344864 is a selective, orally active full agonist of the 5-HT 1F receptor with a high binding affinity (Ki = 6 nM).

**In Vitro and In Vivo Studies**

According to the LY 344864 description, this compound produces an effect similar in magnitude to serotonin itself and possesses the ability to cross the blood-brain barrier. In terms of LY 344864 in vitro activity, the compound binds to the human 5-HT 1F receptor with a Ki of 0.006 μM. It exhibits significantly lower affinity for other serotonin receptors, including 5-HT 1A (0.530 μM), 5-HT 1B (0.549 μM), 5-HT 1D (0.575 μM), and 5-HT 1E (1.415 μM), as well as rat α1- and α2-adrenergic receptors. Additionally, LY 344864 has been identified as an inducer of mitochondrial biogenesis.

Extensive LY 344864 in vivo studies have demonstrated its therapeutic potential across different models. In a rat migraine pain model using male Wistar rats, LY 344864 (0-10 ng/kg; p.o. or i.v.; once) inhibited neurogenic dural inflammation. Specifically, intravenous administration 10 minutes before trigeminal stimulation resulted in an ID50 of 0.6 ng/kg, while oral administration 75 minutes before stimulation yielded an ID50 of 1.2 ng/kg. Furthermore, in a Parkinson’s disease mouse model (male C57BL/6), daily intraperitoneal injections of LY 344864 (2 mg/kg for 14 days) attenuated the loss of tyrosine hydroxylase-immunoreactive (TH-ir) neurons in the striatum and substantia nigra and improved locomotor activity compared to vehicle-treated animals. In conclusion, LY 344864 is a potent and selective 5-HT 1F receptor agonist with promising applications in migraine and Parkinson’s disease research.
